For research published in 2022 study in the journal Food and Chemical Toxicology, scientists examined “the genotoxicity and the intracellular reactive oxygen species induction by physiologically relevant concentrations of three different TiO2 nanomaterials in Caco-2 and HT29-MTX-E12 intestinal cells, while considering the potential influence of the digestion process in the NMs’ physiochemical characteristics.” They found a “DNA-damaging effect dependent on the nanomaterial,” along with the micronucleus assay suggesting “effects on chromosomal integrity, an indicator of cancer risk, in the HT29-MTX-E12 cells, for all the tested TiO2 nanomaterials.” Researchers concluded that the results showcase “evidence of concern” regarding titanium dioxide used as a food additive.

Some websites maintain titanium dioxide is inferior to zinc oxide, another mineral sunscreen ingredient whose core characteristics are similar to those of titanium dioxide. The reality is that titanium dioxide is a great broad-spectrum SPF ingredient and is widely used in all manner of sun-protection products. What gets confusing for some consumers is trying to decipher research that ranks sunscreen ingredients by a UV spectrum graph. By most standards, broad-spectrum coverage for sunscreen ingredients is defined as one that surpasses 360 nanometers (abbreviated as “nm” - how the sun’s rays are measured). Titanium dioxide exceeds this range of protection, but depending on whose research you read, it either performs as well as or slightly below zinc oxide.

Titanium dioxide is typically micronized and coated for use in cosmetics products. The micronizing makes this somewhat heavy-feeling ingredient easier to spread on skin, plus a bit more cosmetically elegant. Micronized titanium dioxide is much more stable and can provide better sun protection than non-micronized titanium dioxide.

Fluorine Chemical, Lithopone 30% CAS No. 1345-05-7, white powder, relative density: 4.136 ~ 4.39 g / mL, insoluble in water. It is a mixture of zinc sulfide and barium sulfate. Inorganic white pigment, widely used in plastics such as polyolefin, vinyl resin, ABS resin, polystyrene, polycarbonate, nylon and polyoxymethylene, and white pigments of paints and inks. It is less effective in polyurethane and amino resins and less suitable in fluoroplastics. It’s true that titanium dioxide does not rank as high for UVA protection as zinc oxide, it ends up being a small difference (think about it like being 10 years old versus 10 years and 3 months old). This is not easily understood in terms of other factors affecting how sunscreen actives perform (such as the base formula), so many, including some dermatologists, assume that zinc oxide is superior to titanium dioxide for UVA protection. When carefully formulated, titanium dioxide provides excellent UVA protection. Its UVA protection peak is lower than that of zinc oxide, but both continue to provide protection throughout the UVA range for the same amount of time.

The production of ROS was studied on white blood cells as a model to screen the effect on eukaryotic cells after being exposed to samples and solar simulated irradiation (according to the level of penetration under the skin). For that purpose, the leukocytes were separated from anticoagulated fresh blood using the Ficoll-Hypaque reactive in a well-known technique [33]. Then, 50 μL of suspensions of P25TiO₂NPs (0.2 mg/mL and 0.02 mg/mL), vitaminB2@P25TiO₂NPs (0.2 mg/mL and 0.02 mg/mL) and vitamin B2 (0.2 mg/mL and 0.02 mg/mL) were prepared and mixed with 50 μL of white blood cells suspension. A solution of 3% H₂O₂ was used as positive control and PBS as negative control. Then, the samples were irradiated using the LED panel for 3 and 6 h to simulate the light penetration into the skin. Also, a set of samples was kept in the dark as control. Finally, the ROS were detected through the colorimetric assay employing the nitroblue tetrazolium salt (NBT salt) and the absorbance at 650 nm was measured. The experiment was reproduced twice; the standard deviation was calculated and p-value < 0.05 were considered significant.
